Transplantationen sind heutzutage eine vorherrschende klinische Lösung zur Behandlung von bspw. Nierenversagen. Dabei ist die Abstoßungsreaktion des Empfängers gegen das Organ noch häufig und bleibt das Hauptproblem schlechthin in diesem Bereich. Ziel des Projektes TRIDIAG ist es, Diagnostik-Tools technologisch und klinisch zu überprüfen, die es ermöglichen, das Risiko von Komplikationen zu messen und damit die Belastung durch diese sehr ernsten Erkrankungen bei Menschen zu lindern.

To this day, transplantation is the main clinical option for the treatment of kidney failure. For example. However, the transplant is often rejected by the host and this is still THE major issue in this field. The aim of the TRIDIAG project is to test the technological and clinical effectiveness of new diagnostic tools capable of measuring the risk of these major complications and therefore ease the burden of these extremely serious effects for patients.

A ce jour, la transplantation rénale est l’option clinique principale pour le traitement - par exemple - des insuffisances rénales. Mais le rejet du greffon par l’hôte est récurrent et reste LA problématique majeure dans ce domaine. Le but du projet TRIDIAG est de valider technologiquement et cliniquement des nouveaux outils diagnostiques permettant de mesurer le risque de développement de ces complications majeures, et donc d''alléger le fardeau de ces événements gravissimes chez l''homme.

Achievements (DE):

Drei Jahre arbeitete das TRIDIAG-Forscherteam aus Basel, Strasbourg, Freiburg, Heidelberg und Mainz daran, ein Diagnostik-Tool für Nieren- und Knochenmarktransplantationen technologisch und klinisch zu validieren, um das Komplikationsrisiko zu messen. Die Rolle von zwei Genen, MICA und MICB, wurde im Bereich der Transplantationsmedizin charakterisiert. Diese Forschung ist für Transplantationen relevant, da diese Gene von Mensch zu Mensch sehr verschieden sind. Die Forscher zeigten insbesondere auf, dass die Kompatibilität zwischen Spendern und Empfängern für diese Gene sehr wichtig ist, um klinische Komplikationen nach einer Knochenmarktransplantation zu vermeiden. Es wurde ein Diagnostiktest entwickelt und validiert, um die Gene in klinischer Routine zu sequenzieren. Die Arbeitsgruppe in Basel hat in dieser Zeit zudem in einer wichtige Studie MICA, MICB und andere ähnliche Moleküle in akuten myeloischen Leukämien untersucht und gezeigt, dass die Suppression dieser Moleküle für das Entkommen von Krebsstammzellen vom Immunsystem wichtig ist. Die enge Zusammenarbeit der beiden Gruppen aus Strasbourg und Basel erlaubte zudem ein neues Gen zu entdecken, welches für schwere angeborene Erkrankungen verantwortlich ist, die durch ausgeprägte Verminderung der weissen Blutkörperchen (Neutropenie) und Infektneigung in Erscheinung tritt. Die Kooperation wird auch nach dem Projektende fortgeführt, indem die Arbeitsgruppen an einer neuen immunologischen Erkrankung forschen.

Achievements (EN):

For three years, the TRIDIAG research team from Basel, Strasbourg, Freiburg, Heidelberg and Mainz worked to technologically and clinically validate a diagnostic tool for kidney and bone marrow transplants in order to measure the risk of complications. The role of two genes, MICA and MICB , has been characterized in the field of transplantation medicine. This research is relevant to transplantation since these genes are very different from person to person. In particular, the researchers showed that the compatibility between donors and recipients for these genes is very important to reduce clinical complications after to avoid bone marrow transplantation. A diagnostic test was developed and validated to sequence the genes in clinical routine. During this time, the working group in Basel also carried out an important study examining MICA, MICB and other similar molecules in acute myeloid leukemias and showed that that the suppression of these molecules for the en escaping cancer stem cells from the immune system is important.
