Cílem této přeshraniční spolupráce je vybudování laboratoře lékařské techniky. To by mělo umožnit nový způsob cíleného výzkumu, jehož záměrem je výroba nosných struktur podobných lidským orgánům v oblasti biotechnologie – se speciálním zaměřením na Tkáňové inženýrství – a navíc vytvoření této laboratoře z úzce spolupracujících institucí by mělo být dlouhodobě udržitelné. Měly by přitom být vyvinuty - pomocí strukturovacích technik na rakouské straně a nanobiologických i makroskopických analytických technik na české straně - modelové systémy, které umožňují zkoumat biologickou tkáň a její chování. Primárním cílem je zde trojrozměrný modelový systém pro cévy, který dovoluje zkoumat mechanismy tvorby cév, reakci na různé substance (např. cholesterol a glukózu) a jejich stimulaci uvnitř cévy. To umožňuje bližší zkoumání vzniku a vývoje arteriosklerózy až po analýzu látkové výměny přes hematoencefalickou bariéru. Plánujeme sledovat reakce uměle vytvořeného cévního systému, který obsahuje biologické buňky, za různých podmínek a na základě tohoto pozorování vytvořit modelový systém pro tvorbu tzv. „Plaques“ (zúžení cév z důvodu celulárních a biomolekulárních usazenin). Tento model dovolí i studium hematoencefalické bariéry v kontextu transportu patogenních virů jako např.: spouštěče klíšťové encefalitidy. Na jedné straně poznatky získané generováním cév v oblasti litografického strukturování a na straně druhé biomolekulární analýza v měřítku nanometru umožní Rakousku a ČR podílet se na jedinečné a slibné progresivní technologii v oblasti bio senzoriky, a sice jako lídr v této oblasti. Vytvořené povrchy mohou v budoucnosti nacházet potenciální uplatnění u mnoha molekulárních čidel (např. těhotenské testy, proteinové čipy), stejně jako při výrobě 3D buněčných struktur, které mohou být využity při stimulované buněčné diferenciaci (umělé orgány, individuální výzkum implantátů a protéz). Dalším důležitým hlediskem je zlepšení pracovních a vzdělávacích podmínek v daném regionu. Ke stanoveným rámcovým podmínkám proto patří perspektiva zahraničního vzdělání a excelentní výzkum, stejně jako opatření usnadňující mobilitu vysoce kvalifikovaných zaměstnanců/zaměstnankyň, studentů/studentek a výměna jejich znalostí.

The aim of this cross-border cooperation is the establishment of a medical technology center. This will enable new and application-oriented research to produce organ-like carrier structures in the field of biotechnology - with a special focus on tissue engineering - and, in the long run, to establish a collaborative network between the institutions involved. Using structuring techniques on the Austrian side, and nanoscopic and macroscopic analysis techniques on the Czech side, model systems will be developed that allow the examination of biological tissue and their behaviour. The first application example is the production of a three-dimensional model system for a blood vessel, which enables research into the buildup mechanisms, the reaction to various substances (e.g. cholesterol and glucose) and their stimulation within a blood vessel. This will enable further research into the emergence and development of arteriosclerosis and serve metabolism analyses across the blood-brain barrier. Monitoring of the artificially created vascular system populated with biological cells is planned, and/or monitoring their reactions to different conditions and creating a model system for the formation of so-called plaques (vascular occlusions due to cellular and biomolecular deposits). The model also allows the study of the blood-brain barrier in view of the transport of pathogenic viruses, e.g. the trigger of tick-borne encephalitis (TBE). The knowledge gained in the field of lithographic structuring in the course of the generation of the vessels on the one hand, and biomolecular analysis at the nanometer scale on the other, offers the potential for Austria and the Czech Republic to participate in a unique and promising future-oriented technology in the field of biosensor technology. Surfaces produced here can be used in a variety of molecular sensors (e.g. pregnancy tests, protein chips, etc), as well as in the manufacture of 3D cell support structures that can be used for stimulated cell differentiation (artificial organs, individualised implant and prosthesis research). Another important focus is the improvement of the working and educational conditions of the participating institutions in the region. Prospects for cross-border education and top-notch research are among the advantages, as well as measures to facilitate the mobility of highly qualified employees and students and their supra-regional exchange of knowledge.

Das Ziel dieser grenzübergreifenden Zusammenarbeit ist der Aufbau eines Medizintechnik-Zentrums. Dies soll neue und anwendungsorientierte Forschung zur Herstellung von organähnlichen Trägerstrukturen auf dem Gebiet der Biotechnologie - mit speziellem Fokus auf Tissue Engineering - ermöglichen und darüber hinaus auf lange Sicht ein Kollaborationsnetzwerk der involvierten Institutionen etablieren. Hierbei sollen mit Hilfe von Strukturierungstechniken auf österreichischer Seite einerseits und nano- und makroskopischen Analysetechniken auf tschechischer Seite andererseits Modellsysteme entwickelt werden, die es erlauben biologisches Gewebe und dessen Verhalten zu erforschen. Das erste Anwendungsbeispiel ist hierbei ein dreidimensionales Modellsystem für ein Blutgefäß zu erzeugen, das die Erforschung der Aufbaumechanismen, der Reaktion auf verschiedene Substanzen (z. B. Cholesterin und Glukose) und deren Stimulation innerhalb eines Blutgefäßes ermöglicht. Dies soll die weitere Erforschung der Entstehung sowie Entwicklung von Arteriosklerose ermöglichen und der Analyse des Stoffwechsels über die Blut-Hirn-Schranke hinweg dienen. Geplant ist hierbei das künstlich geschaffene und mit biologischen Zellen besiedelte Gefäßsystem bzw. dessen Reaktionen auf unterschiedlichen Bedingungen zu überwachen und daraus ein Modellsystem für die Bildung sogenannter Plaques (Gefäßverengungen aufgrund von zellulären und biomolekularen Ablagerungen) zu erstellen. Das Modell erlaubt auch das Studium der Blut-Hirn-Schranke im Hinblick des Transports von pathogenen Viren wie z.B. dem Auslöser der durch Zecken übertragenen FSME-Krankheit. Die im Zuge der Generierung der Gefäße gewonnenen Erkenntnisse auf dem Gebiet der lithographischen Strukturierung einerseits und die biomolekulare Analyse im Nanometer-Maßstab andererseits ermöglichen Österreich und Tschechien an einer einzigartigen und vielversprechenden zukunftsträchtigen Technologie im Bereich der Biosensorik weltweit führend mitzuwirken. Hierbei produzierte Oberflächen finden Anwendung bei einer Vielzahl von molekularen Sensoren (z.B. Schwangerschafts-Tests, Protein-Chips) sowie bei der Herstellung von 3D-Zellträgerstrukturen, die zur stimulierten Zelldifferenzierung genützt werden können (künstliche Organe, individualisierte Implantat- und Prothesenforschung). Ein weiteres wichtiges Augenmerk ist die Verbesserung der Arbeits- und Ausbildungsbedingungen der beteiligten Institutionen in der Region. Perspektiven für grenzübergreifende Ausbildung und exzellente Forschung zählen hierbei ebenso zu den Vorteilen wie Maßnahmen zur Erleichterung der Mobilität hochqualifizierter MitarbeiterInnen und StudentInnen und deren überregionaler Wissensaustausch.

Expected Outputs (CS):

Cílem je vybudovat přeshraniční výzkumné centrum, ve kterém bude možné vyrábět 3D buněčné nosné struktury a mikrofluidní systémy jako modelové systémy. Vybudování tohoto přeshraničního centra přispěje k indikátoru výstupu „Zvýšení počtu pracovišť výzkumu a inovací“. Spolupráce mezi partnery umožní vývoj metod pro výrobu a charakterizaci struktur a mikrofluidních systémů. Úzká spolupráce nezbytná pro úspěšnou realizaci pracovních balíčků 1-4 bude měřena formou společných projektů a publikací v prestižních odborných časopisech stejně jako účastí na konferencích. Příslušné pracovní balíčky tak přispějí ke zvýšení počtu přeshraničních aktivit v oblasti výzkumu a inovací. Vytvoření přeshraničního pracoviště výzkumu a inovací je stejnou měrou definováno realizací investic (API 1-4) a provedením aktivit (APT 1-4).

Das Ziel ist der Aufbau eines grenzübergreifenden Forschungszentrums, in dem 3D-Zellträgerstrukturen und mikrofluidische Systeme als Modellsysteme hergestellt werden können. Der Aufbau dieses grenzübergreifenden Zentrums trägt zum Outputindikator „Erhöhung der Zahl von F&I-Einrichtungen“ bei. Die Kollaboration zwischen den Partnern ermöglicht die Entwicklung von Methoden zur Herstellung und Charakterisierung von Strukturen und mikrofluidischen Systemen. Die enge Zusammenarbeit zur erfolgreichen Umsetzung der Arbeitspakete 1-4 wird in Form von gemeinsamen Projekten und Publikationen in renommierten Fachzeitschriften, als auch Konferenzbesuchen gemessen . Die entsprechenden Arbeitspakete tragen infolgedessen zur Erhöhung der Zahl von grenzübergreifenden Aktivitäten im Bereich F&I bei. Die Etablierung einer grenzübergreifenden F&I Einrichtung definiert sich durch die Umsetzung von Investitionen (API 1-4) und Durchführung von Aktivitäten (APT 1-4) im gleichen Maße.
