Neurodegeneratieve ziekten zoals Parkinson, multiple sclerose en de ziekte van Alzheimer staan bekend voor de intensieve verzorging en daarbij horende ziekenhuisopnames. Methoden voor vroege diagnose en effectieve behandeling zijn dan ook noodzakelijk om de kosten voor gezondheidszorg te verminderen en de levenskwaliteit van patiënten en hun familie te verbeteren. ‘Herinneringen’ neemt de ziekte van Alzheimer als 'proof-of-concept'. Bij ongeveer 5 op 100 families waarin de ziekte vóór het 65ste jaar begint, is de oorzaak meestal erfelijk. Er zijn echter jonge patiënten zonder deze genetische risicofactoren, wat doet vermoeden dat nog ongekende genen en/of externe factoren een rol spelen. De meerderheid ontwikkelt echter symptomen op hogere leeftijd, meestal na het 60ste levensjaar. Dit type Alzheimer kent geen genetische achtergrond. Er wordt aangenomen dat hier sprake is van een accumulatie van veranderingen in het genetisch materiaal door ouderdom en beïnvloedbare risicofactoren zoals chemische stoffen (zware metalen, pesticiden, anæstetica) en levensstijl (zwaarlijvigheid). De mechanismen waarmee deze risicofactoren de verdere ontwikkeling van de ziekte drijven, zullen in ‘Herinneringen’ onderzocht en toegepast worden in nieuwe diagnostische methoden en therapieën. Het project spitst zich onder andere toe op het kunnen stellen van vroege diagnostiek. Zo zal de software van Icometrix, dat oorspronkelijk ontwikkeld werd voor het meten van hersenvervormingen bij multiple sclerosis, aangepast worden voor toepassing op patiënten met Alzheimer. Het principe is dat beelden van MRI-hersenscans omgezet kunnen worden in cijfers, te vergelijken met de cijferwaarden van een bloedtest. Op die manier kunnen meer precieze metingen gebeuren. Deze data worden verder geëvalueerd om de diagnose, opvolging en prognose van Alzheimer te optimaliseren.

Neurodegenerative diseases such as Parkinson's, multiple sclerosis and Alzheimer's disease are known for the intensive care and associated hospitalizations. Early diagnosis and effective treatment methods are therefore necessary to reduce health care costs and improve the quality of life of patients and their families. 'Herinneringen' takes Alzheimer's disease as 'proof-of-concept'. In about 5 out of 100 families in which the disease begins before the age of 65, the cause is usually hereditary. However, there are young patients without these genetic risk factors which would suggest that unprecedented genes and/or external factors play a role. The majority, however, develops symptoms in old age, usually after the age of 60 years old. This type of Alzheimer's does not have a genetic background. It is assumed that there is an accumulation of changes in the genetic material due to age and influencing risk factors such as chemicals (heavy metals, pesticides, anaesthetics) and lifestyle (obesity). The mechanisms by which these risk factors drive the further development of the disease will be investigated in 'Herinneringen' and applied in new diagnostic methods and therapies. The project focuses among other things on being able to make early diagnostics. For example, the Icometrix software, which was originally developed for measuring brain deformations in multiple sclerosis, will be adapted for use on patients with Alzheimer's disease. The principle is that images from MRI brain scans can be converted into numbers, comparable with the numerical values of a blood test. In this way, more precise measurements can be made. These data are further evaluated to optimize the diagnosis, follow-up and prognosis of Alzheimer's disease.
