Im Projekt „CheckNano - Nanosafety Fast Test“ finden sich Partner aus Deutschland und Dänemark zusammen, um einen Prototypen für einen Test zur Identifizierung toxischer Nanopartikel zu entwickeln. Private Unternehmen werden aktiv einbezogen, um den Test zu kommerzialisieren, u.a. durch Spin-Offs. Die deutsch-dänische Region kann damit von der dynamischen Entwicklung auf dem Markt für Nanotechnologie profitieren. Heutzutage werden Nanopartikel (Partikel mit Größen unterhalb 100 nm) in den vielfältigsten Bereichen angewendet, um Produkte radikal zu verbessern, von Kosmetik- und Lebensmittelindustrie zu Medizintechnik und Pharmazie. Nanopartikel durchdringen jedoch unterhalb einer kritischen Größe Zellbarrieren und führen zum Zelltod. Ausführliche Studien der letzten Jahre für Silber-Nanopartikel haben den zugrundeliegenden Mechanismus herausgefunden und diese kritische Größe festgelegt. Eine Kenntnis der Größe und Form der Nanopartikel ist daher unabdingbar für ihre sichere Anwendung und bildet in diesem Projekt die Basis eines innovativen Schnelltestes. Das Projekt kombiniert interdisziplinär in der deutsch-dänischen Grenzregion vorhandene Kompetenzen aus Nanotechnologie (Charakterisierung und Strukturierung), Bio- und Lebensmitteltechnologie und Sensortechnologie. Nano- und Mikrofilter vorselektieren die Partikel, gefolgt von einer exakten optischen Größenbestimmung und einer Feinfiltrierung. Der Schnelltest bietet einen soliden Mehrwert für die Produktsicherheit aller Unternehmen, in denen Nanopartikel in der Produktion zum Einsatz kommen, wie zum Beispiel in der Nahrungsmittel-, der Medizintechnik-, der Kosmetik- und der Pharma- und Verpackungsindustrie. Der Test ist auf Silbernanopartikel als Modellteilchen ausgelegt, da sie aufgrund ihrer antibakteriellen Eigenschaften zu den am häufigsten verwendeten Nanopartikeln in Verbraucherprodukten gehören. Das Verhalten und die Veränderung solcher Partikel in Lebensmitteln ist jedoch wenig bekannt. Da Silber-Nanopartikel in definierten Mengen synthetisiert und physikalisch charakterisiert werden können, sind sie gut geeignet, den Schnelltest für Partikel unterschiedlicher Größe, Form und Oberfläche zu optimieren sowie den Einfluss realer Medien zu untersuchen, wie sie in Lebensmitteln vorkommen.

Description (EN):

The project "CheckNano - Nanosafety Fast Test" combines partners from Germany and Denmark to develop a prototype for a fast test for the identification of toxic nanoparticles. Private companies are actively involved in this project to commercialize the test, e.g. by implementing spin-offs. The German-Danish region can thus benefit from the dynamic development of the nanotechnology market. Today, nanoparticles (particles smaller than 100 nm) are used in a wide range of applications to radically improve products, from the cosmetics and food industries to medical technology and pharmaceuticals. However, nanoparticles penetrate cell barriers below a critical size and lead to cell death. Detailed studies of recent years for silver nanoparticles have identified the underlying mechanism and determined this critical size. Knowledge of the size and shape of nanoparticles is therefore essential for their safe application and forms the basis of an innovative fast test in this project. The project combines interdisciplinary competences from nanotechnology (characterization and structuring), biotechnology, nutrition and sensor technology in the German-Danish border region. Nanofilters and microfilters pre-select the particles, followed by an exact optical sizing and fine filtration. The fast test offers a solid added value for product safety for all companies in which nanoparticles are used in production processes, such as food industries, medical technology industries, cosmetics industries, pharmaceutical and packaging industries. The test is designed for silver nanoparticles as model particles as they are the most commonly used nanoparticles in consumer products due to their antibacterial properties. However, little is known about the behaviour and modification of such particles in food. Since silver nanoparticles can be synthesized and physically characterized in defined quantities, they are well suited to optimize the fast test for particles of different sizes, shapes and surfaces and to investigate the influence of real media as they occur in foodstuffs.

I projektet ""Check Nano - Nano Safety Fast Test"" samler sig partnere fra Tyskland og Danmark til at udvikle en prototype for en test for at identificere giftige nanopartikler. Private virksomheder er aktivt involveret i at markedsføre testen, bl.a. gennem spin-offs. Den dansk-tyske region kan således drage fordel af den dynamiske udvikling på markedet for nanoteknologi. I dag anvendes nanopartikler (partikler med størrelser under 100 nm) i de mest forskellige områder, for at forbedre produkter radikale i kosmetiske og fødevareindustrien ellers medicinteknik og farmaci. Dog penetrerer nanopartikler som er små end en kritisk størrelse celle barrierer og resulterer i celledød. Detaljerede undersøgelser i de senere år for sølv-nanopartikler har opdaget den underliggende mekanisme og defineret denne kritiske størrelse. Kendskab til størrelsen og formen af nanopartiklerne er derfor afgørende for deres sikker anvendelse, og danner i projektet grundlaget for en innovativ kviktest. Projektet kombinerer tværfagligt i den tysk-danske grænseregion eksisterende færdigheder i nanoteknologi (karakterisering og strukturering), bio og fødevareteknologi og sensorteknologi. Nano- og mikrofiltre selekterer partiklerne, efterfulgt af en præcis optisk bestemmelse af størrelser og en fin filtrering. Testet tilvejebringer en betydelig værdi til produktsikkerheden for alle firmaer, som anvender nanopartikler i produktionen, såsom fødevarer, medicin eknik, kosmetik og farmaceutiske og emballageindustrien. Testen er baseret på sølv nanopartikler, da de er blandt de mest anvendte nanopartikler i forbrugerprodukter på grund af deres antibakterielle egenskaber. Adfærd og modifikation af sådanne partikler i fødevarer er dog ikke vel kendt. Eftersom sølvnanopartikler kan syntetiseres i definerede mængder og kan karakteriseres eksakt, er de velegnede til at optimere testet for partikler med forskellig størrelse, form og overflade samt at undersøge indflydelsen af reelle medier, såsom dem der findes i fødevarer.

Achievements (EN):

There are some major findings in this reporting period. The nanoparticles' spectroscopic properties depend heavily on the embedding matrix, as matrix compounds adhere to the particle and form shells. As model nanoparticles for developing a sensor prototype, we used citrate stabilised silver nanoparticles. We investigated these particles' stability under the influence of defined (meat) matrices. The particles' stability is limited, and under certain conditions, we observed half-life times of only minutes. This is an interesting and not necessarily expected finding. The stability of the particles varies on the concentration of the citrate. Changing the solution's PH value causes the particles to disappear within minutes. Pilot studies demonstrated that additional filtering and enrichment solutions for silver nanoparticles are necessary. We managed to fabricate microfluidic units by ourselves with 3D printing technology. We explored field-flow fractionation (FFF) as a filtering method coupled with 3D DLS as optical detection step. By this, we managed to separate NPs ranging up to 100 nanometers from mixtures of NPs of a variety of sizes containing also 200 nm silver particles and bigger polystyrene particles. The final detection method consists of two steps: Step 1: Microfluidic filtering, e.g. 3D printed master moulds and full chips, FFF. Step 2: 1. DLS: 3D DLS and depolarised DLS, to reduce multi-scattering issues as well as minimise biological matrix (background) 2. Step 1 with additional darkfield microscopy as a necessary complementary method. Note that the finding of Ag nanoparticles' instability under real-life conditions (especially in food) significantly affects the nanoparticles' potential toxicity. Both atomic silver and large scale silver aggregates (larger than 100 nm) are not supposed to be dangerous on a cellular level in a real organism – i.e. on cells that are not isolated. It is further noted that detection with DLS is not limited to metallic nanoparticles. This opens up a new door for detecting non-metallic nanoparticles such as nanometric iron oxide or even nano plastics.
