Parmi les filières d’avenir détectées en zone transfrontalière, la santé, les agro-ressources et les biotechnologies sont celles qui détiennent le plus de potentiel de développement. Cependant, le territoire présente une activité en recherche et développement qui est en retrait par rapport aux niveaux nationaux et aux objectifs européens. Afin d’encourager la mise en place d’innovations dans le cadre de ces thématiques émergentes, et face aux besoins de comblement ou de remplacement soulevés par la médecine régénératrice, le projet TEXTOS a pour ambition de proposer aux chirurgiens en orthopédie et en odontologie des substituts de nouvelle génération qui permettront une cicatrisation et une régénération osseuse. L’objectif principal du projet est alors de développer une matrice 3D innovante pour l’ingénierie tissulaire dans le but de reconstruire, régénérer ou remplacer la fonction de tissus ou d’organes déficients. Le prototype sera réalisé en deux temps, avec en premier lieu l’élaboration d’une matrice « cœur » traduisant la preuve de concept, puis celle de la matrice hybride du prototype visé. Le projet TEXTOS permet de regrouper et de fédérer des acteurs de la recherche et de l’innovation de la zone transfrontalière sur des thématiques complémentaires. Il représente une opportunité pour les partenaires de partager leurs pratiques et d’accroitre leur visibilité à l’échelle régionale, nationale et internationale dans le domaine de l’ingénierie tissulaire.

Bij de meest beloftevolle sectoren in de grensstreek hebben gezondheid, agrohulpbronnen en biotechnologieën het grootste ontwikkelingspotentieel. Op het gebied van onderzoek en ontwikkeling hinkt de regio echter achterop in vergelijking met de nationale niveaus en de Europese doelstellingen. Om ontwikkelingen in de context van deze opkomende thema’s aan te moedigen en in het licht van de behoeften aan vulling of vervanging in de regeneratieve geneeskunde wil het project TEXTOS orthopedische en tandchirurgen substituten van de nieuwe generatie aanbieden, die de genezing van wonden en de regeneratie van beenderen bevorderen. Het hoofddoel van het project is daarom een innovatieve 3D-matrix voor weefstelengineering te ontwikkelen voor de reconstructie, regeneratie of vervanging van de weefsels van deficiënte organen. Het prototype zal worden uitgevoerd in twee fasen, met de ontwikkeling van eerst een kernmatrix, die de proof of concept geeft, en vervolgens de hybride matrix van het beoogde prototype. Het project TEXTOS maakt het mogelijk de betrokken partijen bij onderzoek en innovatie in de grensstreek samen te brengen rond complementaire thema’s. Het biedt de partners de kans hun praktijken te delen en hun nationale en internationale zichtbaarheid op het gebied van weefselengineering te verhogen.

Among the promising sectors identified in the cross-border region, health, agro-resources, and biotechnologies are those that have the most potential for development. However, the region is lagging behind in terms of research and development in comparison to national levels and European objectives. In order to encourage the introduction of developments in the context of these emerging themes and faced with the needs for filling or replacement raised by regenerative medicine, the TEXTOS project aims to offer surgeons in orthopaedics and odontology next generation substitutes that will allow wound healing and bone regeneration. The main objective of the project is, therefore, to develop an innovative 3D matrix for tissue engineering with the goal of reconstructing, regenerating, or replacing the tissue of defective organs. The prototype will be realised in two stages, firstly by developing a “core” matrix reflecting the proof of concept, and then the hybrid matrix of the target prototype. The TEXTOS project makes it possible to group and federate research and innovation stakeholders in the cross-border region around complementary themes. It represents an opportunity for the partners to share their practices and to increase their visibility at the national and international level in the field of tissue engineering.

L'objectif général du projet TEXTOS est de développer une matrice 3D innovante pour l'ingénierie tissulaire dans le but de reconstruire, régénérer ou remplacer la fonction de tissus ou d'organes déficients. Le démarrage du projet transfrontalier TEXTOS s'est traduit par des activités de recherche en parallèle avec des actions de communication et de sensibilisation des entreprises de la zone transfrontalière. Des outils de communication ont été mis en place tels que le poster de présentation du projet, une pochette et un Roll-up. Le site internet dédié au projet https://www.textos-interreg.eu/ a été mis en ligne en novembre 2016 et a reçu depuis près de 2000 visites. Le projet a été également publié sur le site ResearchGate qui est un site de communication dédié à la Science et aux scientifiques. A ce jour, notre projet est suivi et vu par plusieurs scientifiques venant d'horizons divers. En effet, Textos continu à susciter l'intérêt auprès de chercheurs avec 20 followers et plus de 425 lectures sur ce portail (https://www.researchgate.net/project/TEXTOS-Development-of-biofunctional-matrix-for-tissue-regeneration). Les partenaires Textos ont réalisé des actions de diffusion et de dissémination transfrontalière auprès du public cible : entreprises industrielles et secteur médical. Dans ce registre, les partenaires ont organisé trois journées techniques dans le domaine biomédical et participé à 11 journées thématiques et 3 salons avec des interventions sous forme de posters et présentations orales. Ainsi, 94 entreprises ont été sensibilisées au projet TEXTOS et un soutien a été apporté à une entreprise de la zone transfrontalière qui a développé un banc innovant pour soigner le mal de dos. Un bilan positif peut être dressé en termes de valorisation auprès de la communauté scientifique avec la participation à 18 conférences et congrès et la publication de 3 articles dans des revues scientifiques internationales. Le point de départ du développement de la matrice 3D étant la réalisation du fil en matériau composite bio-résorbable (PLA-HA) et bio-fonctionnalisé en surface par un revêtement de chitosan. Le choix de matériau substrat (biocomposite PLA-HA), qui constitue la base du fil de la matrice 3D, est guidé par l'application Textos de régénération des tissus osseux. Le partenaire UMons s'est concentré sur la synthèse et la fabrication des composites (PLA/Hydroxyapatite) avec les exigences : homogénéité, aptitude à l'extrusion et modulation des propriétés mécaniques. La problématique d'interface, d'agglomération et de décohésion entre les constituants du composites (PLA et HA), a été traitée avec l'introduction d'une fonctionnalisation des particules de HA avec le (poly)lactide. Ainsi, les propriétés mécaniques du composite ont été améliorées et peuvent être aisément modulées selon l'application Textos. Le partenaire UMons a synthétisé des biocomposites (PLA-HA) en quantité suffisante pour alimenter la fabrication de filaments chez le partenaire Centexbel. L'extrusion des fils biocomposites en grandes quantités sur des machines industrielles a été entamée et permettra de réaliser par la suite de matrices 3D tricotées. L'étape intermédiaire de bio-fonctionnalisation de surface des fils en bio-composites (PLA-HA) consiste à réaliser un dépôt de chitosane afin d'améliorer les propriétés biologiques de la matrice 3D. L'adhésion de ce dépôt sur le fil est assurée par un traitement plasma à pression atmosphérique (PPA) préalable. Un système de traitement de fil en continu a été développé par le CRITT-MDTS combinant deux systèmes de traitement plasma à pression atmosphérique (torche et tube) avec deux systèmes de revêtement (immersion et aspersion). Après avoir mis en place les protocoles d'essais, différentes investigations des propriétés biologiques ont été réalisées depuis les premiers semestres chez le partenaire BIOS de l’URCA. Les résultats marquants sont les suivants : -Démonstration de l'activité antimicrobienne du revêtement de chitosane sur différents substrats de biocomposite (PLA-HA). Aussi, cette étude a montré que le chitosane n'a pas été altéré par le traitement plasma réalisé au CRITT-MDTS. -Des études d'un fil en PLGA commercial ont montré qu'il n'était pas biocompatible de par sa composition chimique où une altération du milieu de culture a été constatée. -Une étude comparative de la morphologie du fil en mono et multi-filaments a montré que dans le cadre du projet TEXTOS, visant plutôt des tissus osseux, une matrice à base de fil en mono-filament est recommandée car elle permet une prolifération des cellules plus modérée que dans le cas d'un multi-filament. - Les membranes réalisées par Centexbel par la technique « electrospinning », structurées en fibres aléatoires, ont été testées par le partenaire BIOS en culture cellulaire. Les résultats obtenus ont montré que les cellules souches adhéraient à la membrane, montrant que le matériau de base PLA-HA est un bon candidat pour la reconstruction osseuse. -Des tests de biocompatibilité sur les biocomposites de TEXTOS (PLA-HA) ont montré un bon comportement des cellules vis à vis des particules d'HA fonctionnalisées et ceci quel que soit la condition étudiée. Comme indiqué dans la littérature, des études ont montré que les nanoparticules, bien qu'elles soient biocompatibles, elles peuvent être phagocytées par les cellules inflammatoires. -La mise au point des modèles de reconstruction osseuse chez le rat. Après obtention des autorisations d'expérimenter sur animaux par le ministère de l'agriculture, des défauts critiques dans les crânes de rat ont été réalisés (40 rats opérés). Les suivis par micro-scanner à 2, 4 et 8 semaines a montré que sur le groupe de rats implantés avec des matériaux à base d'hydoxyapatite, la reconstruction osseuse a été stimulée alors que sur le groupe n'ayant pas reçu d'implantation, les défauts n'ont pas cicatrisés. En outre, la quantité d'os formé est quasi identique à l'os natif. La caractérisation fonctionnelle des tests mécaniques a démontré que la qualité de l'os néoformé peut être améliorée avec les paramètres de tricotage. Enfin, des études in-vivo ont été réalisées : des membranes de collagène fonctionnalisées par les revêtements hybrides (phosphates de Calcium, chitosane et acide hyaluroniques) ont été implantées chez l'animal (défauts sub-critiques de la Calvaria de rats). Le suivi au cours du temps par imagerie chez le petit animal a montré une augmentation significative du tissu osseux régénéré versus les membranes non fonctionnalisées. Les analyses histologiques sur résine montrent la présence d'une matrice collagénique minéralisée, signature de la présence d'un néo-tissu osseux. Les analyses histologiques sur paraffine montrent d'ailleurs la présence d'un tissu vascularisé, synonyme de présence d'un tissu vivant. D’autre part, la quantité d'os formé est quasi identique à l'os natif. Cependant, les propriétés mécaniques obtenues par des essais de nanoindentation sont plus faibles que ceux de l’os natif. Des améliorations peuvent être apportées avec le procédé de tricotage.

The overall objective of the TEXTOS project is to develop an innovative 3D matrix for tissue engineering with the aim of reconstructing, regenerating or replacing the function of deficient tissues or organs. The start of the TEXTOS cross-border project resulted in research activities in parallel with communication and awareness-raising actions among businesses in the cross-border area. Communication tools have been put in place such as the project presentation poster, a cover and a Roll-up. The website dedicated to the project https://www.textos-interreg.eu/ was put online in November 2016 and has received since nearly 2000 visits. The project was also published on ResearchGate, which is a communication site dedicated to science and scientists. To date, our project is followed and seen by several scientists from different backgrounds. Indeed, Textos continues to arouse interest among researchers with 20 followers and more than 425 readings on this portal (https://www.researchgate.net/project/TEXTOS-Development-of-biofunctional-matrix-for-tissue-regeneration). The Textos partners carried out cross-border dissemination and dissemination actions to the target audience: industrial companies and medical sector. In this register, the partners organised three technical days in the biomedical field and participated in 11 thematic days and 3 fairs with interventions in the form of posters and oral presentations. Thus, 94 companies were made aware of the TEXTOS project and support was given to a company in the cross-border area that developed an innovative bench to treat back pain. A positive assessment can be drawn in terms of appreciation to the scientific community with the participation in 18 conferences and congresses and the publication of 3 articles in international scientific journals. The starting point for the development of the 3D matrix is the realisation of the wire in bio-resorbable composite material (PLA-HA) and bio-functionalised on the surface by a chitosan coating. The choice of substrate material (biocomposite PLA-HA), which forms the basis of the 3D matrix wire, is guided by the Textos application of bone tissue regeneration. The partner UMons focused on the synthesis and manufacture of composites (PLA/Hydroxyapatite) with the requirements: homogeneity, extrusion ability and modulation of mechanical properties. The problem of interface, agglomeration and decohesion between the constituents of the composites (PLA and HA), was addressed with the introduction of a functionalisation of HA particles with (poly)lactide. Thus, the mechanical properties of the composite have been improved and can be easily modulated according to the Textos application. The partner UMons synthesised biocomposites (PLA-HA) in sufficient quantity to power filament manufacturing at Centexbel partner. Extrusion of biocomposite wires in large quantities on industrial machines has been initiated and will subsequently make knitted 3D matrices. The intermediate step of surface biofunctionalisation of bio-composite wires (PLA-HA) consists of carrying out a chitosan deposition in order to improve the biological properties of the 3D matrix. The adhesion of this deposit on the wire is ensured by a prior atmospheric pressure plasma (APP) treatment. A continuous wire processing system was developed by CRITT-MDTS combining two atmospheric pressure plasma treatment systems (torch and tube) with two coating systems (immersion and sprinkler). After implementing the test protocols, various investigations of biological properties have been carried out since the first semesters at the BIOS partner of URCA. The key results are as follows: —Demonstration of antimicrobial activity of chitosan coating on different biocomposite substrates (PLA-HA). Also, this study showed that chitosan was not altered by plasma treatment with CRITT-MDTS. Studies of a commercial PLGA wire showed that it was not biocompatible by its chemical composition where alteration of the culture medium was observed. —A comparative study of wire morphology in mono-filament and multi-filament showed that in the TEXTOS project, focusing on bone tissues, a monofilament wire-based matrix is recommended because it allows cell proliferation more moderate than in the case of a multifilament. — The membranes made by Centexbel using the “electrospinning” technique, structured in random fibers, were tested by the partner BIOS in cell culture. The results showed that stem cells adhered to the membrane, showing that the base material PLA-HA is a good candidate for bone reconstruction. Biocompatibility tests on TEXTOS biocomposites (PLA-HA) showed good behavior of cells vis-à-vis functionalised HA particles, regardless of the condition studied. As reported in the literature, studies have shown that nanoparticles, although biocompatible, can be phagocytised by inflammatory cells. —The development of bone reconstruction models in rats. After obtaining authorisations to experiment on animals by the Ministry of Agriculture, critical defects in the rat skulls were made (40 rats operated). Microscanner follow-up at 2, 4 and 8 weeks showed that in the group of rats implanted with hydoxyapatite-based materials, bone reconstruction was stimulated while in the group having not received implantation, the defects did not heal. In addition, the amount of bone formed is almost identical to the native bone. Functional characterisation of mechanical tests has shown that the quality of neoformed bone can be improved with knitting parameters. Finally, in-vivo studies have been carried out: collagen membranes functionalised by hybrid coatings (calcium phosphates, chitosane and hyaluronic acid) were implanted in animals (subcritical defects in rat Calvaria). Time monitoring by imaging in small animals showed a significant increase in regenerated bone tissue versus non-functionalised membranes. Histological analyses on resin show the presence of a mineralised collagenic matrix, signature of the presence of a bone neo-tissue. Histological analyses on paraffin also show the presence of vascularised tissue, synonymous with the presence of a living tissue. On the other hand, the amount of bone formed is almost identical to the native bone. However, the mechanical properties obtained by nanoindentation tests are lower than those of the native bone. Improvements can be made with the knitting process.

Nombre de collaborations transfrontalières encore effectives après la fin du projet : 4. Les différents partenaires entendent poursuivre leur collaboration au-delà des 4 années du projet TEXTOS et du programme Interreg V. Cela se traduirait par l'élaboration d'une thèse de doctorat transfrontalière ou la mise en place de projets de recherche impliquant au moins un partenaire de chaque côté de la frontière sur des programmes internationaux.

Aantal grensoverschrijdende samenwerkingen die ook na afloop van het project worden voortgezet: 4. De verschillende partners willen hun samenwerking ook na de vier jaar van het TEXTOS-project en de Interreg V-programma voortzetten. Dit zal gepaard gaan met het schrijven van een grensoverschrijdende doctoraatsthesis over de implementatie van onderzoeksprojecten, waarbij minstens één partner van elke zijde van de grens in internationale programma’s betrokken is.

Number of cross-border collaborations still in effect after the end of the project: 4. The different partners intend to continue their collaboration beyond the 4 years of the TEXTOS project and the Interreg V programme. This will entail the writing of a cross-border doctoral thesis or the implementation of research projects involving at least one partner from each side of the border in international programmes.

Nombre d’entreprises bénéficiant d’un soutien : 10. Les actions de sensibilisation et de dissémination des résultats menées par les partenaires permettront d’apporter un soutien concret à dix entreprises de la zone transfrontalière. Ce soutien se traduira par une mise en réseau, un conseil ciblé ou un accompagnement orienté selon les besoins de l’entreprise dans les domaines des biotechnologies, du textile ou de la plasturgie. Nombre d’entreprises bénéficiant d’un soutien pour introduire des produits nouveaux pour l’entreprise : 2. Deux de ces entreprises devraient bénéficier d’un soutien pour introduire des nouveaux produits correspondant aux développements prévus dans les deux modules de R&D : « matrice cœur » pour les aspects procédé de fabrication innovant et « matrice hybride » pour une utilisation en régénération tissulaire en milieu médical. Nombre d’établissements de recherche participant à des projets de recherche transfrontaliers : 4. Enfin, tous les partenaires du projet TEXTOS ainsi que les laboratoires universitaires de l’URCA et de l’ UMONS sont considérés comme des centres de recherche participant à des projets de recherche transfrontaliers.

De partners zullen informatiecampagnes voeren en de resultaten van het project verspreiden en op die manier concrete steun leveren aan tien ondernemingen in de grensstreek. Deze steun zal de vorm hebben van netwerken en doelgericht advies of ondersteuning volgens de behoeften van de onderneming op het gebied van biotechnologie, textiel of kunststoffen. Aantal ondernemingen die steun ontvangen om nieuwe producten in de onderneming in te voeren: 2. Twee van deze ondernemingen zouden steun moeten ontvangen om nieuwe producten te introduceren overeenkomstig de ontwikkelingen die de twee O&O-modules voorzien: *kernmatrix* voor de aspecten van innovatieve fabricatieprocessen en hybride matrix voor het gebruik van weefselregeneratie in een medische omgeving. Campaigns designed to raise awareness and disseminate results conducted by the partners will provide concrete support to ten companies in the cross-border region. This support will translate into networking, targeted advice, or focused support according to the needs of the company in the fields of biotechnology, textiles, or plastics. Two of these companies should benefit from support to introduce new products corresponding to the developments foreseen in the two R&D modules: “core matrix” for the innovative manufacturing process aspects and “hybrid matrix” for the use of tissue regeneration in a medical environment.
