Ondanks belangrijke diagnostische en therapeutische vooruitgang blijven cardiovasculaire ongelukken, gelinkt aan ischemische hartziekten de belangrijkste doodsoorzaak in de wereld. De grensoverschrijdende zone tussen Frankrijk en België ontsnapt jammer genoeg niet aan deze realiteit. De ischemische hartziekten vormen de belangrijkste oorzaak van de overlijdens door cardiovasculaire ziekten in de zone (26% bij de mannen en 32% bij de vrouwen). Bijna al deze ziektes hebben een anatomo-pathologische stof gemen, namelijk de aderverkalking. De eerste laesies verschijnen bij elkeen vanaf de kindertijd, nog voor de pubertijd, en ontwikelen zich onverbiddelijk met de leeftijd. Het gevolg zijn klinische manifestaties, niet alleen de progressieve toename van de grootte van het plak, maar eveneens het breken van de plakken. In de klinische praktijk blijkt het primordiaal te zijn de plakken op te sporen die makkelijk breken. Ondanks belangrijke technologische vooruitgang, is de kracht te constateren dat tot op heden geen enkele manier van beeldvorming toelaat om risicovolle plakken te karaktriseren. In deze context wil Nanocardio de systemen voor vroegtijdige diagose van atherosclerose op punt stellen. Het project is gebaseerd op de ontwikkeling van nanoplatformen, uitgerust met magnetische en optische sonden, actief in magnetische (MRI) en optische beeldvorming (fluorescentie). Deze nanoplatformen zullen bovendien uitgerust worden met een specifiek systeem voor de cellulaire herkenning van atheroscletorische plakken. Ons doel is dus de risicovolle atheroscletorische plakken vroegtijdig te detecteren om de kans op hartziekten bij de patiënt te voorkomen en en hem ertegen te beschermen. Het consortium dat hieromtrent samengbracht is, bestaat uit 3 universitaire teams: een Waals team(UMONS), een Frans team(URCA) en een Vlaams team(UGENT), waarvan de knowhow de uitwerking en de karakterisering van beeldvorming met magneto-optische sonden betreftvoor specifieke cellulaire receptoren, aslook de evaluatie van de cytotoxiciteit van deze nanovectoren. Het project steunt op de expertise van twee geassocieerde operatoren, 2 instituten die gespecialisserd zijn in technologie en de miniaturisatie van detectiesystemen(IMEC Leuven), met betrekking tot gezondheidsporblemen (IMEC en IEMN), 2 medische teams (Mons en Reims) en de competitiviteitspolen die gewijd zijn an de gezondheid: Biowin en EuraSanté. Dit zal de banden tussen grensoverschrijdende universitaire teams te versterken.

Les accidents cardiovasculaires liés aux maladies coronariennes restent la principale cause de mortalité et de morbidité à travers le monde. La zone transfrontalière franco-belge n’échappe en effet pas à cette réalité, les cardiopathies constituant l’essentiel des décès cardiovasculaires de la zone (36% chez les hommes et les 24% chez les femmes). Les troubles associés à ces cardiopathies sont liés au développement de l’athérosclérose, une accumulation de lipides, glucides complexes et autres dépôts pouvant boucher les artères et empêcher la circulation sanguine. Le but du projet NanoCardio est de développer une molécule qui, injectée par voie intraveineuse chez le patient, permettra de détecter et d’analyser par IRM et imagerie optique les plaques athéroscléreuses. NanoCardio propose alors de détecter les plaques instables ainsi que leur taille, et de déterminer leur composition chimique. Pour atteindre cet objectif, le projet veut entreprendre le développement de sondes magnéto-optiques ciblant les plaques athéroscléreuses. Il s’agira de nanoparticules équipées d’agents de contraste pour la détection en imagerie, et d’un système de reconnaissance de la plaque. L’approche transfrontalière dans le cadre de ce projet permet de renforcer la collaboration entre des équipes aux savoirs et savoir-faire complémentaires. L’intérêt est aussi de profiter de la proximité géographique entre les parties prenantes pour mener des projets plus ambitieux.

Cardiovascular accidents related to coronary diseases remain the principal cause of mortality and morbidity around the world. The cross-border Franco-Belgian area is not immune to this situation, with heart diseases constituting the bulk of cardiovascular deaths in the area (36% for men and 24% for women). The disorders associated with these heart diseases are related to the development of atherosclerosis, an accumulation of lipids, complex carbohydrates and other deposits that can block the arteries and prevent blood flow. The goal of the NanoCardio project is to develop a molecule that, injected intravenously into the patient, will allow the detection and analysis of atherosclerotic plaques with MRI and optical imaging. NanoCardio thus makes it possible to detect unstable plaques, as well as their size, and to determine their chemical composition. To achieve this objective, the project wants to undertake the development of magneto-optical probes targeting atherosclerotic plaques. It will consist of nanoparticles equipped with contrast agents for imaging detection, and a system of plaque recognition. A cross-border approach within the framework of this project makes it possible to shore up cooperation between teams with complementary knowledge and know-how. The benefit is also to take advantage of the geographic proximity between the stakeholders to conduct more ambitious projects.

Achievements (FR):

Les opérateurs rémois (URCA) et montois (UMONS) du projet NanoCardio (www.nanocardio.eu) ont développé depuis le début du projet les voies de synthèses permettant l'obtention de sondes magnétiques capables d’améliorer la visualisation des vaisseaux sanguins par imagerie par résonance magnétique (IRM). Deux sondes magnétiques, surnommées G1-4GdDO3A et G1-4GdDOTAGA, ont été synthétisées et caractérisées par ces opérateurs. Ces sondes ont ensuite été étudiées sur le petit animal par les opérateurs associés du CHU Ambroise Paré et du Centre de Microscopie et d’Imagerie Moléculaire (CMMI). Ces études ont permis à ces opérateurs d’adapter les conditions d’injections de ces sondes afin d’évaluer leur comportement in vivo et d’obtenir une visualisation optimale des vaisseaux sanguins. De plus, l’UGENT a également participé à l’étude toxicologique de ces sondes, et a démontré leur caractère inoffensif . Suite à ces résultats, les opérateurs rémois et montois du projet ont ensuite voulu coupler ces sondes IRM avec des dérivés fluorescents dans le but d’obtenir un diagnostic renforcé par la combinaison du diagnostic IRM avec une deuxième modalité : l’imagerie optique. Dans ce but, plusieurs systèmes ont été développé (nanoparticules d’oxyde de fer, nanoparticules de silice). La détection de plaques d’athérosclérose aux moyens de ces systèmes a été réalisée par les méthodes d’imagerie, et également par confirmation au moyen de méthodes histologiques. NanoCardio continue également sa démarche de sensibilisation aux maladies cardiovasculaires via son site internet (www.nanocardio.eu) et ses différents réseaux sociaux. Après avoir atteint le cap symbolique des 500 followers sur sa page Facebook, NanoCardio maintient sa communication active dans le domaine général de la santé. La visibilité générale du projet a été également été accrue sur les autres réseaux sociaux (LinkedIn, Twitter) par la publication fréquente de conseils santé ou d’autres rubriques en lien avec les activités du projet. Depuis son début, les opérateurs du projet NanoCardio ont également été actifs dans la communication par leurs participations à de multiples événements, axés soit vers le grand public (Printemps des sciences, Événements Interreg, Mardi des chercheurs), soit vers la communauté scientifique (colloques nationaux et internationaux tels que SFNano, C’Nano, les journées GDR-AIM, les journées de l’EDT-CHIM…).

Achievements (EN):

The NanoCardio (URCA) and Montois (UMONS) operators (www.nanocardio.eu) have developed since the beginning of the project the synthetic pathways allowing the obtaining of magnetic probes capable of improving the visualisation of blood vessels by magnetic resonance imaging (MRI). Two magnetic probes, nicknamed G1-4GdDO3A and G1-4GdDOTAGA, were synthesised and characterised by these operators. These probes were then studied on the small animal by the associated operators of the CHU Ambroise Paré and the Center for Microscopy and Molecular Imaging (CMMI). These studies allowed these operators to adapt the injection conditions of these probes in order to evaluate their in vivo behavior and obtain optimal visualisation of blood vessels. In addition, the UGENT also participated in the toxicological study of these probes, and demonstrated their harmlessness. Following these results, the Rémois and Montois operators of the project then wanted to pair these MRI probes with fluorescent derivatives in order to obtain a diagnosis reinforced by the combination of the MRI diagnosis with a second modality: optical imaging. For this purpose, several systems have been developed (iron oxide nanoparticles, silica nanoparticles). The detection of atherosclerosis plaques by means of these systems was carried out by imaging methods, and also by confirmation using histological methods. Nanocardio also continues to raise awareness of cardiovascular diseases through its website (www.nanocardio.eu) and its various social networks. After reaching the symbolic milestone of the 500 followers on its Facebook page, NanoCardio maintains its active communication in the general field of health. The overall visibility of the project was also increased on other social networks (LinkedIn, Twitter) through frequent publication of health advice or other topics related to the project’s activities. Since its beginning, the operators of the NanoCardio project have also been active in communication through their participation in multiple events, aimed either at the general public (Spring of science, Interreg Events, Tuesday of researchers), or towards the scientific community (national and international colloques such as SFNano, C'Nano, GDR-AIM days, EDT-CHIM days...).

De Franse en Waalse partners UMONS en URCA werken momenteel samen in het kader van een Europees project (Gadolymph). Het project Nanocardio zal ons toelaten deze samenwerking verder te zetten en om ze te versterken dankzij een uitbreiding van het partnerschap door de UGENT. De eerste grensoverschrijdende samenwerking UMONS URCA zal dus uitgebreid worden met een tweede grensoverschrijdende samenwerking met UGENT. De operatoren hopen te profiteren van het hefboomeffect van het NanoCardio-project en het consortium uitgebreid met de geassocieerde operatoren en hun netwerk om Europese oproepen voor projecten aan te vragen.

Les partenaires UMONS et URCA collaborent actuellement dans le cadre d’un projet européen (Gadolymph). Le projet NanoCardio va permettre de poursuivre cette collaboration et de la renforcer grâce à l’extension du partenariat à l’UGENT. La première collaboration transfrontalière UMONS URCA va donc de fait être étendue à une seconde collaboration transfrontalière avec l’UGENT. Les opérateurs espèrent profiter de l’effet de levier du projet NanoCardio et du consortium étendu aux opérateurs associés et à leur réseau pour candidater sur des appels à projets européens.

The partners, UMONS and URCA, are currently collaborating within the framework of a European project (Gadolymph). The NanoCardio project will enable this cooperation to continue and to strengthen it thanks to the extension of the partnership to UGENT. The first cross-border collaboration between UMONS and URCA will thus be extended by a second cross-border collaboration with UGENT. The operators hope to benefit from the leverage effect of the NanoCardio project and of the broader consortium of associated operators and their network to respond to European calls for projects.

Nanocardio heeft de ambitie om in vivo diagnostische systemen voor atherosclerose op punt te stellen. Het heeft als doelstelling om de atherosclerose plaques te visualiseren en hun vroegtijdige actie te analyseren, hun evolutie te voorspellen en de gepaste maatregelen voor de patiënten mogelijk te maken. Nanocardio is er dus op gericht om significant bij te dragen aan een oplossing voor atherosclerose gerelateerde aandoeningen in het grensgebied Frankrijk-België. Vermits het probleem zich mondiaal stelt zullen de resultaten van Nanocardio tevens van grote betekenis zijn buiten de grenszone.

Le projet NanoCardio ambitionne de mettre au point des systèmes in vivo de diagnostic personnalisé de l'athérosclérose. Il a pour but de visualiser les plaques athéroscléreuses et d'analyser leur activité de façon précoce, de façon à prédire leur évolution et permettre une prise en charge adaptée des patients. Il vise donc à répondre à un enjeu de santé transfrontalier majeur, en réduisant l'incidence des problèmes de santé liés à l'athérosclérose sur la population transfrontalière. Comme finalement cet enjeu est un enjeu de santé publique mondial, les résultats du projet Nanocardio pourront également être transposés à cette échelle.

Expected Outputs (EN):

The NanoCardio project aims to develop in vivo systems for the personalised diagnosis of atherosclerosis. Its goal is to view the atherosclerotic plaques and analyse their early-stage activity in such a way as to predict their evolution and adapt the treatment offered to patients. It aims, therefore, at responding to a major cross-border health challenge by reducing the incidence of health problems related to atherosclerosis in the cross-border population. Because this issue is ultimately a global health issue, the results of the NanoCardio project can also be transferred to this level.
