Dans le contexte de l’utilisation de pesticides plus respectueux de l’environnement, les lipopeptides sont des molécules très prometteuses. Les principaux objectifs du projet BIOPROD sont de lever les freins liés à la production industrielle de ces nouveaux biopesticides et des microorganismes producteurs. Ainsi, les actions menées dans ce projet seront : l’optimisation des conditions de production et de purification des molécules en développant, entre autres, des procédés innovants mettant en œuvre des méthodes de screening à haut débit, et le dimensionnement d’installations et la montée en échelle pour parvenir à une future production industrielle des lipopeptides. Ces deux approches techniques contribueront à la détermination d’un coût pour chaque molécule. Ces différentes actions seront complétées par une étude de marché transfrontalière visant à déterminer les pratiques et les attentes des distributeurs de produits phytosanitaires et des agriculteurs. Les résultats seront comparés, afin d’avoir une communication adaptée selon les territoires visant à promouvoir ces nouvelles molécules phytosanitaires, le résultat ultime de ce projet étant la mise sur le marché dans un avenir proche de celles-ci et de permettre ainsi une amélioration de la protection de l’environnement par l’utilisation de nouveaux biopesticides plus respectueux de la nature et des hommes.

As part of the efforts to use more environmentally friendly pesticides, lipopeptides are very promising molecules. The main objectives of the BIOPROD project are to remove the obstacles related to the industrial production of these new biopesticides and their producing microorganisms. Therefore, the actions carried out in this project will be: the optimisation of the conditions for the production and purification of molecules by developing, among other things, innovative processes using high-throughput screening methods, and the sizing of facilities and scaling up to achieve future industrial production of lipopeptides. These two technical approaches will contribute to the determination of a cost for each molecule. These different actions will be complemented by a cross-border market study to determine the practices and expectations of distributors of plant protection products and farmers. The results will be compared, in order to develop communication that is adapted to each territory in order to promote these new phytosanitary molecules, the ultimate result of this project being to bring them onto the market in the near future and thus allow improved environmental protection through the use of new biopesticides that are more respectful of nature and men.

In het kader van de inspanningen voor het gebruik van milieuvriendelijkere pesticiden, zijn lipopeptiden veelbelovende moleculen. De belangrijkste doelstellingen van het project BIOPROD zijn het wegnemen van de belemmeringen voor de industriële productie van deze nieuwe biopesticiden en het feit dat ze micro-organismen produceren. Daarom voorziet dit project de volgende acties: optimalisatie van de voorwaarden voor de productie en zuivering van moleculen, met onder meer de ontwikkeling van innovatieve processen met gebruik van high-throughput screeningmethoden, en de dimensionering van de faciliteiten en de opschaling met het oog op de toekomstige, industriële productie van lipopeptiden. Deze twee technische benaderingen zullen bijdragen tot de bepaling van de kostprijs voor elke molecule. Deze verschillende acties zullen worden aangevuld met een grensoverschrijdende marktstudie om de praktijken en verwachtingen van verdelers van gewasbeschermingsmiddelen en landbouwers te kennen. De resultaten zullen worden vergeleken met het oog op een aan deze regio’s aangepaste communicatie om deze nieuwe fytosanitaire moleculen te promoten. Het einddoel van het project is deze moleculen in de nabije toekomst op de markt te brengen en het milieu beter te beschermen door nieuwe biopesticiden te gebruiken die vriendelijker zijn voor mens en natuur.

L'objectif majeur du projet BIOPROD est de développer de nouvelles stratégies de production et de formulation de biopesticides faiblement toxiques. Les lipopeptides produits par Bacillus subtilis sont des biopesticides très prometteurs mais de nombreux freins restent à lever pour permettre leur utilisation. Le projet BIOPROD comprend ainsi plusieurs axes. Le premier axe consiste à développer de nouveaux procédés innovants permettant d'améliorer les taux de production des lipopeptides tout en réduisant les coûts de production, afin d'envisager la mise en place d'une production à échelle industrielle de ces biomolécules. Dans ce contexte, des travaux ont été menés d'une part pour optimiser les paramètres de production, grâce au génie génétique notamment et d'autre part pour développer des procédés de production innovants. Parmi ces derniers, des travaux menés sur un bioréacteur à film tombant ont permis de mettre en évidence le potentiel de ce type de réacteur pour la surproduction des lipopeptides de Bacillus subtilis. En parallèle, des essais préliminaires menés sur un nouveau type de réacteur, l’airlift, se sont avérés prometteurs pour la production de surfactine et de fengycine. D’autres thématiques sont également en cours d’analyse, avec notamment une étude de l’interaction Bacillus-Trichoderma et de l’impact sur la production de lipopeptides. En parallèle, une montée en échelle du process de production est étudiée. Après la validation de production de mycosubtiline à l'échelle 300 litres, un nouvel essai de 3000 litres a permis de valider les résultats précédemment obtenus. Par ailleurs, en matière de séchage des molécules, pour les 3 lipopeptides, l'atomisation semble être particulièrement efficace et économique. Un second objectif du projet consiste à démontrer l'innocuité des biomolécules, afin de permettre d'envisager leur mise sur le marché. Les essais in vitro indiquent que les lipopeptides non formulés, seuls ou en mélange présentent une cytotoxicité acceptable pour les cellules de lignées humaines. Les premiers essais de biodégradabilité étaient satisfaisants mais ce point doit encore être approfondi. Plusieurs formulations de molécules ou plus directement des micro-organismes sont en cours d'études afin de potentialiser les activités de ces molécules lors de leur application aux champs. L'alginate semble être un matériau prometteur pour réaliser les différentes formulations. En particulier, une bonne stabilité de la concentration cellulaire de Bacillus subtilis dans des billes d’alginate a été mise en évidence après 4 mois de stockage à température ambiante. En parallèle, des essais seront menés afin d'étudier le comportement de ces produits formulés lors de leur pulvérisation sur différentes plantes. Un produit formulé a déjà été validé et d'autres sont à l'étude. Enfin, l'optimisation des procédés de production et le développement d'un produit formulé doit s'accompagner si possible d'une mise sur le marché des biomolécules. Afin d'évaluer cette possibilité, une étude de marché est en cours : plusieurs entreprises cibles en transfrontalier ont été rencontrées afin de déterminer les pratiques et les attentes des distributeurs de produits phytosanitaires. Cette étude permettra une communication adaptée selon les territoires, visant la promotion de ces nouvelles molécules. En se basant sur les retours obtenus, la mise en place d’une formation sur la thématique des biopesticides et les apports de Smartbiocontrol est à l’étude. Ainsi, une amélioration de la protection de l'environnement par l'utilisation de nouveaux biopesticides plus respectueux de la nature et des hommes sera possible. Depuis le début du projet, les premiers résultats scientifiques ont été publiés (6 publications) ou présentés à l'occasion de congrès internationaux (11 présentations orales et 6 posters). Deux conférences de presse, en Belgique et en France, et la publication d'articles dans la presse ont permis de communiquer autour des projets constitutifs du portefeuille SMARTBIOCONTROL (http://www.smartbiocontrol.eu).

Achievements (EN):

The main objective of the BIOPROD project is to develop new strategies for the production and formulation of low toxic biopesticides. The lipopeptides produced by Bacillus subtilis are very promising biopesticides but many brakes remain to be lifted to allow their use. The BIOPROD project thus includes several axes. The first axis is to develop new innovative processes to improve the production rates of lipopeptides while reducing production costs, in order to envisage the establishment of industrial-scale production of these biomolecules. In this context, work has been carried out on the one hand to optimise production parameters, thanks in particular to genetic engineering and on the other hand to develop innovative production processes. Among these, work on a falling film bioreactor has highlighted the potential of this type of reactor for the overproduction of Bacillus subtilis lipopeptides. In parallel, preliminary tests on a new type of reactor, the airlift, proved promising for the production of surfactin and fengycin. Other topics are also being analysed, including a study of the Bacillus-Trichoderma interaction and the impact on lipopeptide production. In parallel, a scale-up of the production process is studied. After the validation of mycosubtiline production at the 300-litre scale, a new 3000-litre test validated the results previously obtained. Moreover, when it comes to the drying of molecules, for the 3 lipopeptides, atomisation seems to be particularly efficient and economical. A second objective of the project is to demonstrate the safety of biomolecules, so that they can be considered for placing on the market. In vitro tests indicate that unformed lipopeptides, alone or in mixtures, have acceptable cytotoxicity for human line cells. The first biodegradability tests were satisfactory, but this point still needs to be further developed. Several formulations of molecules or more directly of microorganisms are being studied in order to potentiate the activities of these molecules when applied to fields. Alginate seems to be a promising material for making different formulations. In particular, a good stability of the cell concentration of Bacillus subtilis in alginate balls was demonstrated after 4 months of storage at room temperature. In parallel, tests will be conducted to study the behaviour of these products formulated during spraying on different plants. One formulated product has already been validated and others are under consideration. Finally, the optimisation of production processes and the development of a formulated product must be accompanied if possible by placing biomolecules on the market. In order to assess this possibility, a market study is ongoing: several cross-border target companies were met to determine the practices and expectations of plant protection distributors. This study will allow a communication adapted according to the territories, aimed at the promotion of these new molecules. Based on the feedback obtained, the implementation of training on the topic of biopesticides and the contributions of SMARTBIOCONTROL is being studied. Thus, an improvement in the protection of the environment through the use of new biopesticides more respectful of nature and humans will be possible. Since the beginning of the project, the first scientific results have been published (6 publications) or presented at international conferences (11 oral presentations and 6 posters). Two press conferences, in Belgium and France, and the publication of articles in the press made it possible to communicate on the projects constituting the SMARTBIOCONTROL portfolio (http://www.smartbiocontrol.eu).
