L'objectif général du projet 3D4MED est de développer des matrices 3D innovantes pour l'ingénierie tissulaire dans le but de reconstruire, régénérer ou remplacer la fonction de tissus ou d'organes déficients. Ces matrices sont un support de croissance développé à partir de polymères biocompatibles avec des propriétés à mémoire forme. L'originalité de ce projet réside d'une part, dans la formulation de nouveaux polymères biocompatibles et d'autre part, dans la préparation d'implants prêt à l'emploi à architectures complexe et individualisés selon un procédé de fabrication additive. Ces implants sont constitués de polymères fonctionnels et biocompatibles de grade GMP avec des propriétés à mémoire de forme, minimisant le traumatisme mécanique pendant et après implantation, tout en favorisant la recolonisation cellulaire et l'intégration tissulaire. Le projet vise à relever plusieurs défis : un défi scientifique et technique visant la réalisation d'un produit innovant ; le second relève de la maturation du produit d'ingénierie tissulaire, en partant du concept vers la réalisation d'un prototype opérationnel validé en condition préopératoire ; une étape clé pour la valorisation du produit auprès des entreprises. Ces défis sont relevés au sein d'un consortium fédérant les différentes expertises complémentaires appartenant à la zone transfrontalière.

Description (EN):

The main objective of the 3D4MED project is to develop an innovative 3D matrix for tissue engineering with the goal of reconstructing, regenerating, or replacing the tissue of defective organs. These matrices are a growth medium developed from biocompatible polymers with shape memory properties. The originality of this project lies, on the one hand, in the formulation of new biocompatible polymers and, on the other hand, in the preparation of ready-to-use implants with complex and individualised architectures using an additive manufacturing process. These implants are made of functional and biocompatible GMP grade polymers with shape memory properties, minimising mechanical trauma during and after implantation, while promoting cell recolonisation and tissue integration. The project aims to meet several challenges: a scientific and technical challenge to produce an innovative product; the second is the further development of the tissue-engineered product, starting from the concept up to the production of an operational prototype validated in preoperative condition; a key step when it comes to promoting the product among companies. These challenges are being met within a consortium comprised of the various complementary areas of expertise found within the cross-border area.

De algemene doelstelling van het project 3D4MED is de ontwikkeling van innovatieve 3D-matrixen voor weefselmanipulatie, om weefsels of organen met verminderde functie te reconstrueren, regenereren of vervangen. Deze matrixen zijn een groeidrager, ontwikkeld op basis van biocompatibele polymeren met vormgeheugeneigenschappen. De originaliteit van dit project ligt enerzijds in de formulering van nieuwe biocompatibele polymeren en anderzijds in de ontwikkeling van gebruiksklare implantaten met ingewikkelde architecturen en geïndividualiseerd volgens een procedé van additieve fabricage. Deze implantaten zijn gemaakt van functionele en biocompatibele GMP-polymeren met vormgeheugen, die het mechanische trauma na implantatie reduceren en celkolonisatie en weefselintegratie bevorderen. Het project staat voor meerdere uitdagingen. Ten eerste is er een wetenschappelijke en technische uitdaging voor de realisatie van een innovatief product. De tweede omvat de rijping van het weefselmanipulatieproduct, van het ontwerp tot de realisatie van een operationeel prototype dat in preoperatieve toestand gevalideerd is, een belangrijke stap in de valorisatie van het product bij de ondernemingen. Deze uitdagingen worden aangegaan door een consortium dat verschillende, complementaire expertises uit de grenszone bundelt.

Achievements (FR):

Les partenaires de 3D4Med ont pu étudier, efficacement, ensemble, les opportunités scientifiques des différentes stratégies qu'ils ont mises en place au début du projet. Pour rappel, 3 études sont encore en cours : (1) l'étude de matériaux préparés par l'UMONS à propriétés piézoioniques (actionneur) en étroite collaboration avec l'UGent, la KULeuven et l'URCA, (2) la régénération tissulaire (tissu dur) et la synthèse de dérivés de gélatine pour la régénération des tissus musculaires (tissus mous) par l'UGent, la KU Leuven et l'URCA et (3) l'utilisation de l'impression 3D pour la régénération osseuse guidée et les supports à mémoire de forme pour le sinus lift avec la collaboration de l'IMT, de l'URCA UMONS et d'Armines . L'UMONS, l'UGent et l'URCA se sont principalement concentrées sur les matériaux à mémoire de forme basés sur des systèmes d'acrylamide (AA), avec des propriétés piézoioniques - c'est-à-dire des actionneurs pour 3D4Med. Après des études comparatives avec plusieurs systèmes à base d'acrylamide, les propriétés de conductivité ionique des matériaux ont été réalisées en collaboration avec le Laboratoire de Physicochimie des Polymères et des Interfaces (LPPI) de l'Université de Cergy-Pontoise (France). Les mesures de fatigue ont montré que les matériaux AA ont un signal très stable sur de nombreux cycles de déformation. L'accent a été mis sur la compréhension des 3 régimes observés dans le signal en fonction de la déformation en suivant la force nécessaire pour appliquer la déformation. Suite à ces premiers résultats, de nouveaux assemblages d'ions empilés ont été envisagés et étudiés. Ces nouveaux assemblages d'ions empilés offrent la possibilité d'adapter l'intensité de la réponse de nos matériaux en fonction de la zone dans laquelle la contrainte est appliquée, permettant une meilleure discrimination de l'emplacement tactile. Les travaux futurs porteront sur le développement d'implants à libération contrôlée. Concernant les travaux sur la régénération du tissu musculaire, la KULeuven et l'UGent ont pu travailler sur la pré-vascularisation du renforcement musculaire. Le projet a développé une étude sur la pré-vascularisation en ingénierie vers un réseau tubulaire 3D complexe. Les tissus biologiques ont été perfusés avec un système vasculaire et le profil de biocompatibilité d'un nouvel hydrogel électroactif à base de méthacryloyle de gélatine (GelMA) contenant le polymère électroactif poly (3,4-éthylènedioxythiophène):poly(styrènesulfonate) (PEDOT:PSS) a été étudié. Cet hydrogel permettrait d'obtenir un matériau aux propriétés de conduction électrique intéressantes combinées à des propriétés de biodégradabilité et de biocompatibilité, contrairement à celles classiquement utilisées. D'autre part, dans le cadre de l'étude des tissus durs, un nouveau dérivé de gélatine a également été développé à l'UGent permettant l'incorporation de groupements réticulables supplémentaires. Il en résulte un module de compression plus élevé et un gonflement plus faible après réticulation, ce qui rend le matériau plus adapté à la différenciation des cellules souches dans la voie ostéogénique. Le troisième axe développé sur la régénération osseuse a été étudié par l'IMT en étroite collaboration avec l'URCA. À ce stade du projet, des mélanges de polyuréthane/polyester extrudés à différents ratios de polyester ont été étudiés. Certains des mélanges présentent des propriétés de mémoire de forme intéressantes. Des études de relation entre la structure (MEB), des tests de mémoire de forme (tests de traction à 37°C), de dégradation dans le temps en milieu biologique (DSC, SEC) ont permis d'identifier l'intérêt de ce type de matériau pour nos applications médicales (sinus lift). Il est intéressant de noter que des travaux sur l'intérêt d'utiliser l'hydroxyapatite dans des matrices polyester pour la régénération osseuse guidée ont également montré qu'il est possible de préparer des supports à forme contrôlée par impression 3D. Ces supports 3D sont biocompatibles, bioactifs et biodégradables et possèdent éventuellement des propriétés de mémoire de forme intéressantes pour notre application.

Achievements (EN):

The partners of 3D4Med were able to study, effectively, together, the scientific opportunities of the different strategies they put in place at the beginning of the project. As a reminder, 3 studies are still ongoing: (1) the study of materials prepared by UMONS with piezoonic properties (accelerator) in close collaboration with the UGent, KULeuven and URCA, (2) tissue regeneration (hard tissue) and synthesis of gelatin derivatives for muscle tissue regeneration (soft tissue) by UGent, KU Leuven and URCA, and (3) the use of 3D printing for guided bone regeneration and sinus lift memory media in collaboration with IMT, UMONS URCA and Armines. UMONS, UGent and URCA focused mainly on form memory materials based on acrylamide (AA) systems, with piezoonic properties — i.e. actuators for 3D4Med. After comparative studies with several acrylamide-based systems, the ion conductivity properties of materials were carried out in collaboration with the Laboratory of Physicochemistry of Polymers and Interfaces (LPPI) of the University of Cergy-Pontoise (France). Fatigue measurements have shown that AA materials have a very stable signal over many deformation cycles. Emphasis was placed on understanding the 3 regimes observed in the signal as a function of deformation following the force necessary to apply the deformation. Following these first results, new assemblages of stacked ions were considered and studied. These new stacked ion assemblies offer the possibility to adapt the intensity of the response of our materials according to the area in which the stress is applied, allowing better discrimination of the tactile location. Future work will focus on the development of controlled-release implants. Regarding the work on muscle tissue regeneration, KULeuven and UGent were able to work on the pre-vascularisation of muscle building. The project developed a study on pre-vascularisation in engineering towards a complex 3D tubular network. The biological tissues were perfused with a vascular system and the biocompatibility profile of a new electroactive hydrogel based on gelatin methacryloyl (GelMA) containing the electroactive polymer poly (3,4-ethylenedioxythiophene):poly(Styrenesulfonate) (PEDOT:PSS) was studied. This hydrogel would make it possible to obtain a material with interesting electrical conduction properties combined with biodegradability and biocompatibility properties, unlike those conventionally used. On the other hand, as part of the study of hard tissues, a new gelatin derivative was also developed at UGent allowing the incorporation of additional reticulable groups. This results in a higher compression module and lower swelling after crosslinking, making the material more suitable for the differentiation of stem cells in the osteogenic pathway. The third axis developed on bone regeneration was studied by IMT in close collaboration with URCA. At this stage of the project, mixtures of polyurethane/polyester extruded at different polyester ratios have been studied. Some of the mixtures have interesting shape memory properties. Structural relationship studies (MEB), form memory tests (37 °C tensile tests), biological degradation over time (DSC, SEC) have made it possible to identify the interest of this type of material for our medical applications (sinus lift). It is interesting to note that work on the value of using hydroxyapatite in polyester matrices for guided bone regeneration has also shown that it is possible to prepare 3D-controlled media. These 3D media are biocompatible, bioactive and biodegradable and possibly have interesting form memory properties for our application.

Pour développer de nouveaux dispositifs médicaux par impression 3D, l'équipe mettra en œuvre des actions transfrontalières de sensibilisation, recherche appliquée, dissémination, accompagnement et soutien auprès des industriels. Les partenaires poursuivront leur collaboration au-delà du programme Interreg pour pérenniser le réseau transfrontalier créé.

Expected Results (EN):

To develop new medical devices via 3D printing, the team will roll out cross-border awareness campaigns, applied research, dissemination, support and assistance to manufacturers. The partners will continue their collaboration beyond the Interreg programme to secure the long-term future of the cross-border network thus set up.

Om nieuwe medische hulpmiddelen te ontwikkelen met 3D-printing zal het team gebruikmaken van grensoverschrijdende sensibiliseringsacties, toegepast onderzoek, verspreiding, begeleiding en steun aan de producenten. De partners zullen hun samenwerking ook na het Interreg-programma verderzetten om het gevormde grensoverschrijdende netwerk een blijvend karakter te geven.

Expected Outputs (FR):

Plusieurs acteurs économiques sont susceptibles d'être intéressés par le projet : au moins 5 entreprises en hauts-de-France, 8 en Wallonie et 5 en Flandres. Deux d'entre elles devraient bénéficier d'un soutien spécifique pour introduire de nouveaux produits. Le projet ambitionne de réaliser un transfert industriel de chaque côté de la frontière. En amont, d'autres fabricants pourraient bénéficier des retombées de la mise sur le marché. Dix publication scientifiques sont visées, ainsi que des participations à des conférences, covnentions d'affaires et salons.

Expected Outputs (EN):

Several economic stakeholders are likely to be interested in the project: at least 5 companies in the Hauts-de-France region , 8 in Wallonia and 5 in Flanders. Two of them should receive specific support to introduce new products. The project targets industrial transfer on each side of the border. Upstream, other manufacturers could benefit from the spin-offs of its marketing. Ten scientific publications are targeted, as well as participation in conferences, business conventions and trade fairs.

Meerdere economische actoren kunnen belangstelling tonen voor het project: minstens 5 ondernemingen in Hauts-de-France, 8 en Wallonië en 5 in Vlaanderen. Twee van hen zouden een specifieke steun moeten krijgen om de nieuwe producten te introduceren. Het project heeft de ambitie een industriële overdracht aan beide zijden van de grens te verwezenlijken. Later zouden ook andere fabrikanten baat kunnen hebben bij de introductie op de markt. Er worden tien wetenschappelijke publicaties beoogd, alsook deelname aan conferenties, zakenconventies en salons.
